"How much does the room cost?" is correct.
The tense used, is the present simple, and we only add "s" to the verb in the affirmative, NOT the negative or questions.
For example:
"How much does a room cost?
It costs £100 a night. But….
It doesn't cost £50 a night.
I hope this is helpful for your English, it is a frequent error in English.
